Adult Dosing
- Erosive esophagitis: 20 mg orally once daily for 4 to 8 weeks
- Gastric ulcer: 20 mg orally once daily for up to 8 weeks
Pediatric Dosing
- Safety and efficacy in pediatric patients under 18 years have not been established
Indications
- Treatment of erosive esophagitis
- Treatment of gastric ulcer
Mechanism of Action
- Potassium-competitive acid blocker (P-CAB)
- Reversibly inhibits the H+/K+-ATPase pump in gastric parietal cells, blocking gastric acid secretion
Contraindications
- Hypersensitivity to zastaprazan or any component of the formulation
- Co-administration with rilpivirine-containing products
Adverse Reactions
- Nausea
- Diarrhea
- Headache
- Nasopharyngitis
- Elevated liver enzymes
Drug Interactions
- Drugs dependent on gastric pH for absorption (e.g., ketoconazole, itraconazole, iron salts): absorption may be significantly reduced
- CYP3A4 substrates, inhibitors, or inducers may alter plasma concentrations
