A02BD17
Forms & Strengths
- Vonoprazan 20 mg / Amoxicillin 500 mg co-packaged therapy
Adult Dosing

- Administered with or without food
Pediatric Dosing
- Safety and effectiveness in pediatric patients have not been established
Indications
- Treatment of Helicobacter pylori infection in adults
Mechanism of Action
- Vonoprazan is a potassium-competitive acid blocker (P-CAB) that inhibits H+/K+-ATPase in the gastric parietal cells
- Amoxicillin is a bactericidal beta-lactam penicillin antibiotic that inhibits bacterial cell wall synthesis
Contraindications
- Known hypersensitivity to vonoprazan, amoxicillin, or any penicillin class antibiotic
- History of severe hypersensitivity reactions (e.g., anaphylaxis, Stevens-Johnson syndrome) to beta-lactams
Adverse Reactions
- Diarrhea
- Dysgeusia
- Vaginal candidiasis
- Abdominal pain
- Headache
- Hypertension
Drug Interactions
- Drugs dependent on gastric pH for absorption (e.g., ketoconazole, iron salts)
- Probenecid decreases renal tubular secretion of amoxicillin, increasing its blood levels
- May decrease efficacy of oral contraceptives due to altered gut flora affecting absorption
- Methotrexate clearance may be reduced by amoxicillin, increasing risk of toxicity
