Forms & Strengths
- oral suspension
- oral tablets: 50mg, 100mg
- injectable solution: 50mg/mL
Adult Dosing
- 100-200mg three times daily for liver conditions
- 50-100mg twice daily for biliary disorders
- dose adjustment may be necessary for hepatic or renal impairment
Pediatric Dosing
- 20-50mg twice daily for children > 6 years with biliary atresia
- 10-20mg twice daily for children < 6 years with liver disease
- pediatric dosing must be individualized and titrated according to clinical response
Indications
- treatment of chronic liver diseases
- biliary atresia in children
- primary biliary cirrhosis
- cholestasis
Mechanism of Action
- stimulation of bile production and bile flow
- enhancement of hepatocellular transport and secretion
- inhibition of bile salt-induced hepatocyte damage
Contraindications
- known hypersensitivity to cyclobutyrol
- acute or severe liver decompensation
- pregnancy or breastfeeding unless benefits outweigh risks
Adverse Reactions
- gastrointestinal disturbances: nausea, diarrhea, abdominal pain
- allergic reactions: rash, urticaria, pruritus
- elevations in liver enzymes, potentially indicating hepatotoxicity
Drug Interactions
- CYP3A4 and CYP2C9 inhibitors may increase cyclobutyrol plasma levels
- inducers of CYP3A4 may decrease cyclobutyrol efficacy
- enhanced sedative effects when combined with central nervous system depressants
- cyclobutyrol may increase INR when co-administered with warfarin
