Forms & Strengths
- Extended-release tablets: 75 mg
Adult Dosing
- 75 mg orally once daily, taken mid-morning
Pediatric Dosing
- Safety and efficacy not established in children under 12 years
- Not recommended for pediatric use
Indications
- Short-term adjunct in the management of exogenous obesity
Mechanism of Action
- Sympathomimetic amine with pharmacologic activity similar to amphetamines
- Stimulates the release of norepinephrine and dopamine from nerve terminals in the central nervous system
- Suppresses appetite via the satiety center in the hypothalamus
Contraindications
- History of cardiovascular disease (e.g., arrhythmias, severe hypertension, coronary artery disease)
- Advanced arteriosclerosis
- Glaucoma
- Hyperthyroidism
- Known hypersensitivity to sympathomimetic amines
- History of drug abuse
- Concomitant use of monoamine oxidase inhibitors (MAOIs) or within 14 days of discontinuation
- Agitated states
Adverse Reactions
- Insomnia
- Tachycardia
- Palpitations
- Elevated blood pressure
- Nervousness, anxiety, or restlessness
- Dry mouth
- Constipation or diarrhea
- Dizziness or headache
Drug Interactions
- Monoamine oxidase inhibitors (MAOIs): risk of hypertensive crisis
- General anesthetics: risk of cardiac arrhythmias
- Other central nervous system stimulants: additive stimulation and increased toxicity
- Antihypertensive agents: efficacy may be diminished by sympathomimetic activity
- Insulin and oral hypoglycemic agents: dosage requirements may be altered
