Forms & Strengths

Adult Dosing
- 40 mg orally once daily in the morning
- Duration of treatment typically limited to a few weeks
Pediatric Dosing
- Safety and efficacy not established in pediatric patients
- Not recommended for use in children
Indications
- Short-term adjunctive treatment of exogenous obesity
Mechanism of Action
- Indirect sympathomimetic amine
- Acts as a prodrug for amphetamine
- Stimulates the release of norepinephrine and dopamine from central storage sites
- Suppresses appetite via the satiety center in the hypothalamus
Contraindications
- Known hypersensitivity to sympathomimetic amines
- History of cardiovascular disease (e.g., severe hypertension, arrhythmias)
- Advanced arteriosclerosis
- Glaucoma
- History of drug abuse or dependence
- Concurrent treatment with monoamine oxidase inhibitors (MAOIs) or within 14 days of discontinuation
- Hyperthyroidism
- Agitated states

- Palpitations
- Elevated blood pressure
- Dry mouth
- Nervousness
- Dizziness
- Tremor
- Psychotic episodes (with prolonged use or abuse)
- Dependence and tolerance
Drug Interactions
- Monoamine oxidase inhibitors (MAOIs): severe hypertensive crisis risk
- General anesthetics: risk of cardiac arrhythmias
- Other sympathomimetic agents: additive cardiovascular effects
- Antihypertensives: antagonism of hypotensive effect
- Tricyclic antidepressants: potentiation of cardiovascular effects
