Forms & Strengths
- Oral pellets: 10 grams per packet
Adult Dosing
- Hereditary orotic aciduria: 1 gram/kg orally four times daily
- 5-Fluorouracil or capecitabine overdose/toxicity: 10 grams orally every 6 hours for 20 doses
Pediatric Dosing
- Hereditary orotic aciduria (pediatric patients): 1 gram/kg orally four times daily, adjusted based on clinical response and growth
- 5-Fluorouracil or capecitabine overdose/toxicity (pediatric patients): 10 grams orally every 6 hours for 20 doses (if body surface area supports dosing or per specific protocol)
Indications
- Treatment of hereditary orotic aciduria
- Emergency treatment of patients following a 5-fluorouracil or capecitabine overdose
- Management of early-onset severe or life-threatening toxicity within 96 hours following 5-fluorouracil or capecitabine administration
Mechanism of Action
- Acts as a pyrimidine analog acting as a substrate source for uridine
- Competes with 5-fluorouracil (5-FU) metabolites to prevent incorporation into RNA and DNA and mitigate cellular toxicity
- Bypasses the enzymatic block in pyrimidine synthesis in hereditary orotic aciduria
Contraindications
- Known hypersensitivity to uridine triacetate or its components
Drug Interactions
- No clinically significant pharmacokinetic drug interactions identified
- May decrease the efficacy of 5-fluorouracil or capecitabine if administered concurrently (intended for toxicity reversal)
