doxecitine and doxribtimine
A16AX29
Forms & Strengths
- Oral pellets: doxecitine 29 mg / doxribtimine 18 mg
- Oral pellets: doxecitine 58 mg / doxribtimine 36 mg
Adult Dosing
- Thymidine kinase 2 (TK2) deficiency: Weight-based dosing, typically administered 3 to 4 times daily
- Refer to specific protocol guidelines based on total daily dose requirements and patient weight
Pediatric Dosing
- Thymidine kinase 2 (TK2) deficiency in pediatric patients: Weight-based dosing administered 3 to 4 times daily
- Adjust dosage regularly as the child's weight changes to maintain therapeutic levels
Indications
- Treatment of pediatric and adult patients with confirmed thymidine kinase 2 (TK2) deficiency
Mechanism of Action
- Combines deoxynucleosides (doxecitine and doxribtimine) to bypass the enzymatic defect in mitochondrial DNA synthesis caused by TK2 deficiency
- Restores intracellular pool of deoxynucleotides to improve mitochondrial function and mitigate myopathic symptoms
Contraindications
- History of severe hypersensitivity to doxecitine, doxribtimine, or any component of the formulation
Adverse Reactions
- Diarrhea
- Abdominal pain
- Nausea and vomiting
- Elevated liver transaminases
Drug Interactions
- No formal clinical drug-drug interaction studies have been performed
- Potential for interactions with drugs sharing mitochondrial toxicity profiles or nucleoside transporters
