Forms & Strengths
- Lyophilized powder for reconstitution: 1000 mg
- Lyophilized powder for reconstitution: 4000 mg
- Lyophilized powder for reconstitution: 5000 mg
Adult Dosing
- 60 mg/kg intravenous once weekly
- Administer at a rate not to exceed 0.08 mL/kg/min
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Chronic augmentation therapy in patients with congenital alpha-1 antitrypsin deficiency and clinical evidence of emphysema
Mechanism of Action
- Augments low levels of alpha-1 antitrypsin in blood and lung tissue
- Inhibits neutrophil elastase to protect lung connective tissue from enzymatic degradation
Contraindications
- History of severe systemic hypersensitivity reaction to alpha-1 antitrypsin
- IgA deficiency with antibodies against IgA due to risk of severe anaphylaxis
Adverse Reactions
- Headache
- Dizziness
- Dyspnea
- Nausea
- Urticaria
- Pruritus
- Infusion-related reactions (fever, chills)
Drug Interactions
- No formal drug interaction studies have been conducted
- Live vaccines should be used with caution in immunocompromised states, though direct interactions are not documented
