Forms & Strengths
- oral tablets: 500mg
- topical cream: 0.5%
- topical ointment: 0.5%
- injectable solution: 5000mg/vial
Adult Dosing
- PO: 1-2g q8h for 2 days before surgery
- Topical: apply 2-4 times daily
- IV: 8-12mg/kg/day, divided into 2-4 doses
Pediatric Dosing
- PO: 30-50mg/kg/day, divided into 2-4 doses for children >1 year
- PO: 30mg/kg/day, divided into 2-4 doses for infants 6-12 months
- Topical: apply 2-4 times daily for children >3 months
Indications
- decreases bacterial load preoperatively in abdomen
- treats infections caused by susceptible organisms
- used as adjunct to Hepatitis A prophylaxis
Mechanism of Action
- inhibits protein synthesis by binding to 30S ribosomal subunit
- interferes with initiation complex of peptide formation
- bactericidal
Contraindications
- hypersensitivity to neomycin or other aminoglycosides
- intestinal obstruction
- myasthenia gravis
- pregnancy
Adverse Reactions
- OTC: vertigo, tinnitus, hearing loss
- GI: nausea, vomiting, diarrhea
- allergic reactions: rash, pruritus
Drug Interactions
- increased risk of nephrotoxicity and OTC with diuretics
- increased risk of neuromuscular blockade with succinylcholine
- increased absorption when given with urea
