Forms & Strengths
- Oral tablets 100 mg
- Intravenous injection solution 50 mg/5 mL
Adult Dosing
- Oral: 200 mg to 300 mg daily administered in 3 divided doses
- Intravenous: 300 mg to 400 mg daily infused over several hours for acute ischemic conditions
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Angina pectoris prophylaxis and treatment
- Management of coronary artery disease
- Prevention of restenosis following percutaneous transluminal coronary angioplasty (PTCA)
Mechanism of Action
- Inhibits cyclic nucleotide phosphodiesterase, increasing intracellular cAMP levels
- Acts as a potent coronary vasodilator
- Inhibits platelet aggregation and platelet-derived growth factor (PDGF) release
Contraindications
- Hypersensitivity to trapidil or any formulation excipients
- Recent myocardial infarction with severe hemodynamic instability
- Severe hypotension or cardiogenic shock
- Active hemorrhagic disorders
Adverse Reactions
- Gastrointestinal distress, nausea, and vomiting
- Dizziness, headache, and flushing
- Hypotension and tachycardia
- Bleeding diathesis secondary to antiplatelet activity
Drug Interactions
- Increased risk of bleeding when co-administered with anticoagulants or antiplatelet agents
- Enhanced hypotensive effects when used concurrently with other vasodilators or antihypertensives
