Forms & Strengths
- Tablets: 25mg
- Tablets: 50mg
Adult Dosing
- Initial dose: 25mg orally once daily
- Target dose: 50mg orally once daily after 4 weeks
Pediatric Dosing
- Safety and efficacy not established in pediatric patients
Indications
- Treatment of resistant hypertension
- Chronic kidney disease with hypertension
Mechanism of Action
- Directly blocks vasopressin V1A receptors
- Reduces peripheral resistance, increases cardiac output
- Activates endothelial nitric oxide synthase to increase NO production
Contraindications
- Hypersensitivity to aprocitentan or any component
- Pregnancy
Adverse Reactions
- Hypotension
- Hyperkalemia
- Anemia
- Dizziness
- Fatigue
- Headache
- Nausea
Drug Interactions
- Dual blockade of the renin-angiotensin-aldosterone system
- Other agents that increase potassium levels
- Cytochrome P450 3A4 inhibitors or inducers
