Forms & Strengths
- Oral capsules: 200 mg
- Oral tablets: 400 mg
Adult Dosing
- Peripheral vascular disease: 400 mg orally 4 times daily
- Cerebrovascular insufficiency: 200 to 400 mg orally 4 times daily
- Maintenance dose: Taper to lowest effective dose after initial response (typically 400 mg twice daily)
Pediatric Dosing
- Safety and efficacy not established in pediatric patients
- Not recommended for use in children
Indications
- Adjunctive management of intermittent claudication in peripheral vascular disease
- Adjunctive treatment of chronic cerebrovascular insufficiency
Mechanism of Action
- Direct relaxant effect on vascular smooth muscle, primarily on peripheral and cerebral vessels
- Weak papaverine-like antispasmodic activity
- May inhibit phosphodiesterase, increasing intracellular cyclic AMP and promoting vasodilation
Contraindications
- Hypersensitivity to cyclandelate or any component of the formulation
- Acute myocardial infarction
- Severe hypotension
- Recent cerebral hemorrhage
Adverse Reactions
- Transient flushing of the face and neck
- Mild gastrointestinal distress (pyrosis, nausea, eructation)
- Headache and dizziness
- Tachycardia or palpitations at higher doses
Drug Interactions
- Additive hypotensive effects with antihypertensive agents and vasodilators
- Potential enhancement of other CNS depressants or alcohol effects
