Forms & Strengths
- Capsules: 10mg
- Oral solution: Not commercially available
Adult Dosing
- Initial dose: 10mg orally once daily
- Maintenance dose: 20-40mg orally twice daily
- Maximum dose: 100mg daily
- Dose titration: every 2-3 days
Pediatric Dosing
- Children: not established
- Infants: contraindicated
Indications
- Pheochromocytoma
- Hypertension due to pheochromocytoma
- Hypertensive emergencies
Mechanism of Action
- Nonselective alpha-adrenergic receptor antagonist
- Blocks alpha-1 and alpha-2 adrenergic receptors
- Causes vasodilation and decreased peripheral resistance
Contraindications
- Coronary insufficiency
- Angina pectoris
- Heart failure
- Infants and newborns
Adverse Reactions
- Orthostatic hypotension
- Dizziness
- Sodium retention
- Tachycardia
- Gastrointestinal disturbances
Drug Interactions
- Tricyclic antidepressants: increased hypotension
- Antihistamines: additive sedation
- Monoamine oxidase inhibitors: increased hypotension
- Nitrates: increased hypotension
