Forms & Strengths
- Tablets: 150mg, 300mg
- Injectable solution: 100mg/5mL, 200mg/10mL
- Suppositories: 150mg
Adult Dosing
- Peripheral vascular disease: 450-600mg/day orally in 2-3 divided doses
- Cerebrovascular disease: 300-450mg/day orally in 2 divided doses
- Parenteral administration: 200-400mg/day IV in 2-4 divided doses
Pediatric Dosing
- No established pediatric dosing; use with caution in children under 18 years old
- Limited data available in pediatric population
Indications
- Treatment of peripheral vascular disease (e.g. intermittent claudication, Raynaud's phenomenon)
- Cerebrovascular disease (e.g. stroke, transient ischemic attacks)
- Diabetic angiopathy
Mechanism of Action
- Potent calcium channel blocker
- Alpha-adrenergic receptor antagonist
- Inhibits platelet aggregation
- Dilates peripheral blood vessels, increasing blood flow
Contraindications
- Hypersensitivity to buflomedil or other calcium channel blockers
- Severe heart failure, cardiogenic shock
- Unstable angina, acute myocardial infarction
Adverse Reactions
- Headache, dizziness, lightheadedness
- Nausea, vomiting, gastrointestinal disturbances
- Hypotension, orthostatic hypotension
- Rash, pruritus, allergic reactions
Drug Interactions
- Concomitant use with other vasodilators, beta blockers, or calcium channel blockers may increase hypotensive effects
- Interacts with grapefruit juice, increasing buflomedil bioavailability
- May interact with CYP3A4 inhibitors (e.g. ketoconazole, erythromycin)
