Adult Dosing
- Peripheral vascular disorders: 25 to 50 mg orally three times daily
- Titrate dose based on clinical response and tolerance (maximum 150 mg daily)
Pediatric Dosing
- Safety and efficacy in pediatric patients not established
Indications
- Peripheral vascular diseases characterized by vasospasm (e.g., Raynaud's syndrome, acrocyanosis)
Mechanism of Action
- Competitive alpha-adrenergic receptor antagonist
- Produces peripheral vasodilation by blocking postsynaptic alpha-1 and alpha-2 adrenergic receptors
- Reduces total peripheral resistance and increases blood flow to the skin and extremities
Contraindications
- Hypersensitivity to azapetine or other benzazepine derivatives
- Conditions where vasodilation is hazardous (e.g., recent myocardial infarction, severe coronary artery disease)
- Hypotension
Adverse Reactions
- Postural hypotension
- Dizziness, vertigo, and syncope
- Nausea, vomiting, and diarrhea
- Nasal congestion
- Tachycardia and palpitations
Drug Interactions
- Antihypertensive agents: Enhanced hypotensive effect
- Vasodilators: Increased risk of severe hypotension
- Sympathomimetics: May antagonize the vasodilatory effects of azapetine
