Forms & Strengths
- Intravenous powder for solution: 10 mg
Adult Dosing
- 5 mg/hour administered as a continuous intravenous infusion
- Initiate treatment as soon as possible after aneurysmal subarachnoid hemorrhage (aSAH)
- Continue treatment for up to 14 days or until the risk of vasospasm has passed
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Prevention of vasospasm-related morbidity and mortality after aneurysmal subarachnoid hemorrhage (aSAH)
Mechanism of Action
- Endothelin receptor antagonist (ERA) with high selectivity for the endothelin A (ETA) receptor
- Inhibits endothelin-1-induced vasoconstriction, thereby reducing the incidence of cerebral vasospasm and secondary ischemic events following aSAH
Contraindications
- Hypersensitivity to clazosentan or any of its excipients
- Pregnancy
Adverse Reactions
- Hypotension
- Anemia or decreased hemoglobin
- Edema and fluid retention
- Lung infection or pneumonia
- Headache
Drug Interactions
- Strong CYP3A4 inducers may decrease clazosentan exposure
- Strong CYP3A4 inhibitors may increase clazosentan exposure
- Concomitant administration with other vasodilators may increase the risk of severe hypotension
