Forms & Strengths
- Oral tablets: 20 mg
- Oral tablets: 50 mg
Adult Dosing
- Angina pectoris: 50 mg to 100 mg orally two to three times daily
- Maximum daily dose: 300 mg orally
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
- Not recommended for use in children
Indications
- Prophylaxis and treatment of chronic stable angina pectoris
Mechanism of Action
- Non-selective calcium channel blocker
- Inhibits transmembrane influx of calcium ions into cardiac muscle and vascular smooth muscle
- Decreases myocardial oxygen demand by reducing peripheral vascular resistance and afterload
Contraindications
- Hypersensitivity to fendiline or any component of the formulation
- Sick sinus syndrome or second- or third-degree AV block without a pacemaker
- Cardiogenic shock
- Manifest heart failure
- History of QT prolongation or concurrent use of drugs that prolong the QT interval
Adverse Reactions
- Dizziness
- Headache
- Flushing
- Peripheral edema
- Nausea
- Fatigue
- QT interval prolongation and associated ventricular arrhythmias (torsades de pointes)
Drug Interactions
- Other antihypertensive agents or vasodilators (additive hypotensive effect)
- Beta-blockers or digitalis glycosides (risk of bradycardia and AV conduction disturbances)
- Drugs known to prolong the QT interval (increased risk of severe arrhythmias)
- CYP3A4 inhibitors (potential for increased plasma concentrations of fendiline)
