volanesorsen
C10AX18
Forms & Strengths
- Subcutaneous injection: 284 mg/1.5 mL in a single-dose pre-filled syringe
Adult Dosing
- 284 mg subcutaneously once weekly
Pediatric Dosing
- Safety and efficacy not established in pediatric patients
Indications
- Adjunct to diet for treatment of familial chylomicronaemia syndrome (FCS) in patients with high risk for pancreatitis
Mechanism of Action
- Antisense oligonucleotide (ASO) inhibitor targeting hepatic apolipoprotein C-III (ApoC-III) mRNA
- Reduces synthesis of ApoC-III, leading to decreased VLDL, reduced triglyceride levels, and enhanced clearance of triglyceride-rich lipoproteins via LPL-dependent and independent pathways
Contraindications
- History of severe thrombocytopenia related to volanesorsen
- Platelet count below baseline normal limits (<140,000/mcL)
- Hypersensitivity to volanesorsen or any excipients
Adverse Reactions
- Severe thrombocytopenia
- Bleeding diathesis
- Injection site reactions (erythema, pain, pruritus)
- Flu-like symptoms (pyrexia, fatigue, chills, headache)
- Nausea
Drug Interactions
- Antiplatelet agents and anticoagulants (increased risk of bleeding when combined with volanesorsen-induced thrombocytopenia)
- Drugs associated with thrombocytopenia (additive risk of severe platelet reduction)
