Forms & Strengths
- Oral tablets: 0.5 mg, 1 mg
Adult Dosing
- Initial: 0.5 mg to 1 mg daily in divided doses
- Maintenance: Up to 2 mg to 3 mg daily adjusted based on clinical response and thyroid function tests
Pediatric Dosing
- Safety and efficacy in pediatric patients not established
Indications
- Treatment of resistance to thyroid hormone (RTH)
- Adjunct in specific metabolic and dermatological conditions requiring thyroid receptor activation
Mechanism of Action
- Thyroid hormone receptor agonist (primarily acts on thyroid hormone receptor beta)
- Suppresses TSH secretion via negative feedback at the pituitary level, helping overcome peripheral tissue resistance
Contraindications
- Hypersensitivity to tiratricol or excipients
- Untreated adrenal insufficiency
- Acute myocardial infarction and severe cardiovascular disease
- Thyrotoxicosis

Drug Interactions
- Oral anticoagulants: May potentiate the hypoprothrombinemic effect
- Antidiabetic agents: May decrease glycemic control, requiring dosage adjustment of insulin or oral hypoglycemics
- Cholestyramine/Colestipol: Decrease absorption of tiratricol
- Sympathomimetics: Increased risk of cardiovascular toxicity
