Forms & Strengths
- Promestriene is available as tablets, 2.5mg
- Tablets are also available in 5mg strength
Adult Dosing
- For menopausal symptoms, the usual dose is 2.5mg daily
- For menstrual disorders, the dose can range from 2.5 to 5mg daily
Pediatric Dosing
- Safety and efficacy have not been established in pediatric patients
- Promestriene is not recommended for children
Indications
- Treatment of menopausal symptoms
- Treatment of menstrual disorders such as dysmenorrhea and metrorrhagia
- Treatment of endometriosis
Mechanism of Action
- Promestriene acts as an estrogen receptor agonist
- It binds to estrogen receptors in target tissues, inducing estrogenic effects
Contraindications
- Pregnancy and lactation
- History of thromboembolic disorders
- Known or suspected estrogen-dependent tumors
Adverse Reactions
- Nausea and vomiting
- Headache and dizziness
- Breast tenderness and vaginal discharge
- Menstrual irregularities
Drug Interactions
- Inducers of CYP3A4 may decrease promestriene levels
- Inhibitors of CYP3A4 may increase promestriene levels
- Concomitant use with other estrogens may increase the risk of adverse effects
