Forms & Strengths
- Trimegestone 0.1mg and estradiol 1mg tablets
- Trimegestone 0.5mg and estradiol 1mg tablets
Adult Dosing
- 1 tablet orally once daily
- Cycling regimen: 28-day pack, 24 days of hormone tablets, 4 days of placebo tablets
Pediatric Dosing
- Not indicated for use in children prior to menarche
- Use in adolescent females only when clearly indicated
Indications
- Treatment of moderate to severe vasomotor symptoms due to menopause
- Treatment of moderate to severe symptoms of vulvar and vaginal atrophy due to menopause
Mechanism of Action
- Trimegestone is a progestin that acts by binding to the progesterone receptor
- Estradiol is an estrogen that acts by binding to the estrogen receptor
- Combination of trimegestone and estradiol helps to maintain the health and function of female reproductive tissues
Contraindications
- Known or suspected pregnancy
- Known or suspected breast cancer
- Active deep vein thrombosis, pulmonary embolism, or history of these conditions
- Active or recent arterial thromboembolic disease
- Liver dysfunction or disease
- Known or suspected estrogen-dependent neoplasia
Adverse Reactions
- Nausea
- Breast tenderness
- Headache
- Vaginal bleeding or spotting
- Weight changes
- Mood changes
Drug Interactions
- Inducers of CYP3A4 may decrease the levels of trimegestone and estradiol
- Inhibitors of CYP3A4 may increase the levels of trimegestone and estradiol
- May interact with other medications, including antihypertensives, antidiabetics, and anticoagulants
