evocalcet
H05BX06
Forms & Strengths
- Oral tablets: 1 mg
- Oral tablets: 2 mg
- Oral tablets: 4 mg
Adult Dosing
- Initial dose: 1 mg to 2 mg orally once daily
- Maintenance dose: Titrated based on serum calcium and intact PTH levels, up to a maximum of 8 mg once daily
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Treatment of secondary hyperparathyroidism in patients on hemodialysis
Mechanism of Action
- Calcimimetic agent that allosterically modulates the calcium-sensing receptor (CaSR) on parathyroid cells
- Increases the sensitivity of the CaSR to extracellular calcium
- Suppresses parathyroid hormone (PTH) secretion and lowers serum calcium and phosphorus levels
Contraindications
- Hypersensitivity to evocalcet or any of its excipients
- Patients with baseline serum calcium levels below the lower limit of normal
Adverse Reactions
- Hypocalcemia
- Nausea
- Vomiting
- Diarrhea
- Abdominal pain
- Decreased appetite
- QT prolongation secondary to hypocalcemia
Drug Interactions
- CYP3A4 inhibitors may increase systemic exposure of evocalcet
- CYP3A4 inducers may decrease systemic exposure of evocalcet
- Concomitant administration with other calcium-lowering drugs increases the risk of severe hypocalcemia
