Adult Dosing
- Recurrent vulvovaginal candidiasis (RVVC): Day 1, 600 mg (administered as three 196 mg capsules taken as a single dose) divided into two doses separated by 12 hours (e.g., two capsules in the morning, one capsule in the evening); Day 2, 600 mg similarly divided. Maintenance: 145 mg once weekly starting 14 days after the initial loading doses for 11 weeks (total duration 12 weeks)
Pediatric Dosing
- Safety and efficacy in pediatric patients below the age of 18 years have not been established
Indications
- Reduction in the incidence of recurrent vulvovaginal candidiasis (RVVC) in females with a history of RVVC who are not of reproductive potential
Mechanism of Action
- Inhibitor of fungal sterol 14-alpha-demethylase (CYP51), which is necessary for fungal ergosterol biosynthesis, leading to accumulation of toxic methylated sterols and fungal cell death
Contraindications
- Females of reproductive potential
- Pregnant or lactating women
- History of hypersensitivity to oteseconazole
Drug Interactions
- Strong BCRP inhibitors: increased exposure and potential toxicity of oteseconazole
- BCRP substrates (e.g., rosuvastatin): increased exposure and potential adverse effects of the substrate drug
