Forms & Strengths
- Tablets: 300mg zidovudine, 150mg lamivudine, 300mg abacavir
- Tablets: 300mg zidovudine, 300mg lamivudine, 600mg abacavir
Adult Dosing
- 1 tablet twice daily
- Dose adjustment for renal impairment: CrCl < 50 mL/min, not recommended
Pediatric Dosing
- Children weighing 25kg or more: 1 tablet twice daily
- Children weighing 20-24kg: 0.5 to 1 tablet twice daily, depending on formulation
- Dose adjustment for pediatric renal impairment: CrCl < 50 mL/min, not recommended
Indications
- Treatment of HIV-1 infection in combination with other antiretrovirals
- Recommended for use in adults and children weighing at least 25kg
Mechanism of Action
- Zidovudine, lamivudine, and abacavir are nucleoside reverse transcriptase inhibitors (NRTIs)
- Inhibit HIV replication by incorporating into viral DNA and terminating chain elongation
Contraindications
- Hypersensitivity to zidovudine, lamivudine, or abacavir
- Presence of HLA-B*5701 allele due to risk of abacavir hypersensitivity
Adverse Reactions
- Common: headache, nausea, fatigue, diarrhea, rash
- Serious: lactic acidosis, liver damage, severe skin reactions, abacavir hypersensitivity
Drug Interactions
- Concomitant use with emtricitabine: not recommended due to duplication of lamivudine component
- Concomitant use with stavudine or zalcitabine: not recommended due to increased risk of toxicity
