Forms & Strengths
- Tablets: 300 mg lamivudine, 300 mg tenofovir disoproxil, 100 mg doravirine
- Fixed-dose combination
Adult Dosing
- 1 tablet orally once daily
- No dosage adjustment needed for renal impairment with CrCl ≥50 mL/min
- Not recommended for patients with severe renal impairment (CrCl <30 mL/min)
Pediatric Dosing
- Safety and efficacy not established in pediatric patients <18 years
- Not recommended for children due to lack of data
Indications
- Treatment of HIV-1 infection in adults without prior antiretroviral treatment experience
- HIV-1 infection in certain patients with prior antiretroviral treatment experience
Mechanism of Action
- Lamivudine: nucleoside reverse transcriptase inhibitor (NRTI)
- Tenofovir disoproxil: NRTI
- Doravirine: non-nucleoside reverse transcriptase inhibitor (NNRTI)
Contraindications
- Hypersensitivity to lamivudine, tenofovir disoproxil, doravirine, or any component of the formulation
- Concomitant use with certain medications (e.g., dofetilide)
Adverse Reactions
- Most common: headache, nausea, fatigue, diarrhea
- Less common: renal impairment, bone effects (e.g., osteomalacia), immune reconstitution syndrome
Drug Interactions
- Concomitant use with rifampin, rifabutin, rifapentine, St. John's Wort, or carbamazepine may decrease doravirine concentrations
- Not recommended with certain medications (e.g., dofetilide, ivabradine) due to increased risk of adverse reactions
