Adult Dosing
- Herpes zoster: 400 mg orally once daily after a meal for 7 days
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Treatment of herpes zoster
Mechanism of Action
- Helicase-primase inhibitor; inhibits viral DNA replication by binding to the complex of viral proteins encoded by genes UL5, UL8, and UL52
Contraindications
- Hypersensitivity to amenamevir or any component of the formulation
- Concomitant use with strong CYP3A inducers (e.g., rifampin)
Adverse Reactions
- Nasopharyngitis
- Blood bilirubin increased
- Alanine aminotransferase (ALT) increased
- Aspartate aminotransferase (AST) increased
- Headache
Drug Interactions
- CYP3A substrates: May alter plasma concentrations of sensitive substrates due to weak induction/inhibition
- Strong CYP3A inducers: Decreases amenamevir exposure, reducing efficacy
- OATP1B1/3 substrates: Potential for altered pharmacokinetics due to transporter interactions
