Forms & Strengths
- Suspension for intramuscular injection: rVSV-ZEBOV (Ervebo), single-dose vial containing 1 mL (at least 2 x 10^7 plaque-forming units)
- Heterologous 2-dose regimen: Ad26.ZEBOV (Zabdeno) 0.5 mL suspension for injection AND MVA-BN-Filo (Mvabea) 0.5 mL suspension for injection
Adult Dosing
- rVSV-ZEBOV: Single 1 mL intramuscular dose
- Ad26.ZEBOV and MVA-BN-Filo regimen: Dose 1 is Ad26.ZEBOV (0.5 mL IM), followed by Dose 2 as MVA-BN-Filo (0.5 mL IM) administered approximately 8 weeks later
Pediatric Dosing
- rVSV-ZEBOV: Indicated for individuals 1 year of age and older; single 1 mL IM dose
- Ad26.ZEBOV and MVA-BN-Filo: Indicated for individuals 1 year of age and older; Ad26.ZEBOV 0.5 mL IM followed by MVA-BN-Filo 0.5 mL IM 8 weeks later
Indications
- Active immunization for the prevention of disease caused by Zaire ebolavirus in individuals 1 year of age and older
- Pre-exposure prophylaxis for individuals at high risk of exposure, such as healthcare workers and outbreak responders
Mechanism of Action
- Recombinant, replication-competent vesicular stomatitis virus expressing the Zaire ebolavirus glycoprotein (rVSV-ZEBOV) induces humoral and cellular immune responses against the Ebola virus surface glycoprotein
- Heterologous prime-boost regimen (Ad26.ZEBOV and MVA-BN-Filo) uses modified adenovirus and poxvirus vectors encoding Ebola virus glycoproteins to stimulate robust, long-term humoral and cell-mediated immunity
Contraindications
- History of a severe allergic reaction (e.g., anaphylaxis) to any component of the vaccines
- Severe immunodeficiency (for live-attenuated rVSV-ZEBOV vaccine)
Adverse Reactions
- Injection site pain, swelling, and erythema
- Fever, fatigue, headache, myalgia, and arthralgia
- Lymphadenopathy
- Rare hypersensitivity and anaphylactic reactions
Drug Interactions
- Immunosuppressive therapies may reduce the immune response to the vaccines
- Administration with other vaccines should be separated by at least 4 weeks unless co-administration is specifically validated
- Prophylactic antipyretics may blunt the immune response if given around the time of vaccination
