Forms & Strengths
- Injection: 100mg, 200mg, 500mg, 1000mg
- Capsule: 50mg
Adult Dosing
- Hodgkin lymphoma: 150mg/m2 IV days 1 and 15 of 28-day cycle
- Melanoma: 250mg/m2 IV days 1-5, repeat every 3 weeks
- Soft tissue sarcoma: 250mg/m2 IV days 1-5, repeat every 3 weeks
Pediatric Dosing
- Limited data available, use with caution in pediatric patients
Indications
- Hodgkin lymphoma
- Malignant melanoma
- Soft tissue sarcoma
- Neuroblastoma
Mechanism of Action
- Alkylates DNA and RNA, inhibiting DNA replication and transcription
- Forms methylated bases which miscode during DNA replication
Contraindications
- Hypersensitivity to dacarbazine
- Severe liver or kidney impairment
Adverse Reactions
- Myelosuppression
- Nausea, vomiting, diarrhea
- Fatigue, anorexia, weight loss
- Cardiotoxicity
- Hepatotoxicity
Drug Interactions
- Phenytoin: decreases phenytoin levels
- Imipramine: decreases imipramine levels
- Cimetidine: increases dacarbazine toxicity
- Digoxin: increases digoxin levels
