Forms & Strengths
- Powder for injection, 2 mg and 4 mg vials
- Lyophilized powder for reconstitution
Adult Dosing
- 3 mg/m² IV every 3 weeks
- Adjust dose based on toxicity and creatinine clearance
Pediatric Dosing
- Safety and efficacy not established in pediatric patients
- Use not recommended in pediatric patients
Indications
- Advanced colorectal cancer
- Palliative treatment of colorectal cancer
Mechanism of Action
- Inhibits thymidylate synthase
- Interferes with DNA synthesis
- Inhibits cell division
Contraindications
- Hypersensitivity to raltitrexed
- Severe renal impairment (creatinine clearance <20 mL/min)
Adverse Reactions
- Neutropenia
- Thrombocytopenia
- Anemia
- Diarrhea
- Nausea
- Vomiting
- Fatigue
Drug Interactions
- Increases toxicity with 5-fluorouracil
- Decreases efficacy with folic acid or folinic acid
- Nephrotoxic agents increase raltitrexed toxicity
