Forms & Strengths
- Lyophilized powder for injection: 1 mg/vial, 5 mg/vial
- Solution for injection: 2 mg/mL
- Solution for infusion: 1 mg/mL, 2 mg/mL
Adult Dosing
- Dosage for non-small cell lung cancer: 3.0-4.0 mg/m^2 IV, every 1-2 weeks
- Dosage for breast cancer: 3.0-4.0 mg/m^2 IV, every 1-2 weeks
- Dosage for lymphomas and leukemias: 2.5-3.0 mg/m^2 IV, every 1-2 weeks
Pediatric Dosing
- Dosage for pediatric cancer: 2-3 mg/m^2 IV, every 1-2 weeks
- Dose adjustment according to tolerance and response
Indications
- Treatment of non-small cell lung cancer
- Treatment of breast cancer
- Treatment of lymphomas and leukemias
- Treatment of malignant melanoma
Mechanism of Action
- Interferes with microtubule assembly, causing cell cycle arrest at metaphase
- Inhibits mitotic spindle formation, leading to apoptosis
Contraindications
- Hypersensitivity to vindesine or other vinca alkaloids
- Pregnancy and lactation
- Severe bone marrow depression
Adverse Reactions
- Myelosuppression, neutropenia, thrombocytopenia, anemia
- Neurotoxicity, peripheral neuropathy, CNS depression
- Nausea, vomiting, diarrhea, alopecia
Drug Interactions
- Enhanced toxicity with concurrent use of CYP3A4 inhibitors
- Reduced efficacy with concurrent use of CYP3A4 inducers
- Caution with concurrent use of liver function tests-altering agents
