Forms & Strengths
- 45mg/mL solution for injection in 6.3mL or 12.6mL vials
- Pegylated long-acting form of irinotecan
Adult Dosing
- Initial dose: 112mg/m² every 14 days
- Dose adjustments based on individual tolerability and adverse reactions
- Maximum dose: 150mg/m² every 14 days
Pediatric Dosing
- Not established for patients under 18 years old
- Safety and efficacy in pediatric patients not evaluated
Indications
- Treatment of metastatic breast cancer
- Treatment of ovarian cancer
- Treatment of colorectal cancer
Mechanism of Action
- Inhibits topoisomerase I, an enzyme involved in DNA replication
- Induces DNA damage, leading to cell death
Contraindications
- Hypersensitivity to irinotecan or any component of etirinotecan pegol
- Severe neutropenia
- Diarrhea that is severe or persistent
Adverse Reactions
- Neutropenia
- Diarrhea
- Fatigue
- Nausea and vomiting
- Alopecia
Drug Interactions
- CYP3A4 and CYP3A5 inducers (e.g., rifampin) decrease etirinotecan exposure
- CYP3A4 and CYP3A5 inhibitors (e.g., ketoconazole) increase etirinotecan exposure
- Co-administration with atazanavir, a strong CYP3A inhibitor, is not recommended
