binimetinib
L01EE03
Adult Dosing
- 45 mg orally twice daily approximately 12 hours apart
- Continue until disease progression or unacceptable toxicity
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Unresectable or metastatic melanoma with BRAF V600E or V600K mutation, in combination with encorafenib
Mechanism of Action
- Reversible inhibitor of mitogen-activated extracellular signal-regulated kinase 1 and 2 (MEK1/2) kinase activity
- Inhibits the MAPK/ERK pathway, suppressing proliferation in BRAF-mutant tumor cells
Contraindications
- Known hypersensitivity to binimetinib or any of its components
Adverse Reactions
- Fatigue
- Nausea
- Vomiting
- Diarrhea
- Abdominal pain
- Visual disturbances (serous retinopathy, retinal detachment)
- Cardiomyopathy (decreased ejection fraction)
- Venous thromboembolism
- Creatine phosphokinase (CPK) elevation
Drug Interactions
- Avoid concurrent strong CYP3A4 inducers which may decrease binimetinib exposure
- Minimal potential for pharmacokinetic drug interactions via CYP enzymes or transporters
