Forms & Strengths
- Parsaclisib is available as 10mg and 20mg tablets
- Also available as 50mg and 100mg capsules for oral administration
Adult Dosing
- Recommended dose for B cell malignancies is 20mg orally once daily
- Dose reduction to 10mg is recommended for patients with severe renal impairment
Pediatric Dosing
- Safety and efficacy have not been established in pediatric patients
- Dosing for pediatric patients has not been determined due to lack of clinical data
Indications
- Treatment of adult patients with relapsed or refractory follicular lymphoma
- Treatment of adult patients with relapsed or refractory marginal zone lymphoma
- Treatment of adult patients with relapsed or refractory mantle cell lymphoma
Mechanism of Action
- Parsaclisib is a selective PI3K inhibitor that targets the PI3K alpha, delta, gamma isoforms
- Inhibits PI3K signaling pathway, resulting in decreased cell proliferation and increased cell death in malignant B cells
Contraindications
- Hypersensitivity to parsaclisib or any of its components
- Severe hepatic impairment
Adverse Reactions
- Common adverse reactions include fatigue, diarrhea, nausea, pyrexia, and cough
- Serious adverse reactions include pneumonia, neutropenia, and thrombocytopenia
- Increased risk of infections, including severe and fatal infections
Drug Interactions
- Avoid concomitant use of strong CYP3A inhibitors
- Concomitant use of moderate CYP3A inhibitors may require dose reduction
- Parsaclisib may increase concentrations of CYP3A substrates
