Forms & Strengths
- Lyophilized powder for injection 100mg/vial
- Single-dose vial for reconstitution
Adult Dosing
- 5.4mg/kg IV every 3 weeks
- Administer as an intravenous infusion over 90 minutes
Pediatric Dosing
- Not established for patients under 18 years
- Limited data available for pediatric use
Indications
- Unresectable or metastatic HER2-positive breast cancer
- Unresectable or metastatic HER2-positive gastric cancer
- Previously treated HER2-low breast cancer
Mechanism of Action
- Topoisomerase I inhibitorpayload released upon binding to HER2
- Induces DNA damage and apoptosis in cancer cells
- Targets HER2-expressing cells via monoclonal antibody
Contraindications
- Hypersensitivity to trastuzumab deruxtecan
- Pregnancy and lactation due to potential fetal harm
Adverse Reactions
- Interstitial lung disease/pneumonitis
- Neutropenia
- Anemia
- Fatigue
- Nausea
- Decreased appetite
Drug Interactions
- Avoid concomitant use with other topoisomerase inhibitors
- Monitor for increased risk of myelosuppression with other myelosuppressive agents
