Forms & Strengths
- Intravenous infusion: 100 mg/4 mL single-dose vial
- Intravenous infusion: 400 mg/16 mL single-dose vial
Adult Dosing
- Metastatic colorectal cancer: 5 mg/kg every 2 weeks or 7.5 mg/kg every 3 weeks with infusional 5-FU-based therapy
- Non-small cell lung cancer: 15 mg/kg every 3 weeks with carboplatin/paclitaxel
- Glioblastoma: 10 mg/kg every 2 weeks
- Metastatic renal cell carcinoma: 10 mg/kg every 3 weeks with interferon alfa
- Cervical cancer: 15 mg/kg every 3 weeks with paclitaxel and cisplatin or topotecan
- Epithelial ovarian, fallopian tube, or primary peritoneal cancer: 15 mg/kg every 3 weeks with chemotherapy
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Metastatic colorectal cancer
- Non-squamous non-small cell lung cancer
- Recurrent glioblastoma
- Metastatic renal cell carcinoma
- Persistent, recurrent, or metastatic cervical cancer
- Epithelial ovarian, fallopian tube, or primary peritoneal cancer
Mechanism of Action
- Recombinant humanized monoclonal IgG1 antibody that binds to human vascular endothelial growth factor (VEGF)
- Inhibits the binding of VEGF to its receptors (Flk-1 and Flt-1) on endothelial cells
- Reduces microvascular growth and inhibits metastatic angiogenesis
Contraindications
- Severe hypersensitivity to bevacizumab or any of its components
- Inadequately controlled hypertension
- Recent history of severe hemoptysis
Adverse Reactions
- Epistaxis and hypertension
- Gastrointestinal perforation
- Thromboembolic events (arterial and venous)
- Proteinuria and impaired wound healing
- Hemorrhage and posterior reversible encephalopathy syndrome (PRES)
Drug Interactions
- Increased risk of severe diarrhea and leukopenia when combined with irinotecan-containing regimens
- Increased risk of hemorrhage with anticoagulants and antiplatelet agents
- Enhanced cardiovascular toxicity when combined with anthracyclines or prior radiation
