Forms & Strengths
- 4.5 mg vial for reconstitution
Adult Dosing
- 3 mg/m^2 on days 1, 4, and 7 for newly diagnosed AML
- 3 mg/m^2 on days 1, 4, and 7 for relapsed or refractory AML
Pediatric Dosing
- 0.1 mg/kg on days 1, 4, and 7 for relapsed or refractory AML
Indications
- Newly diagnosed acute myeloid leukemia (AML)
- Relapsed or refractory AML
Mechanism of Action
- Antibody-drug conjugate targeting CD33
- Delivers calicheamicin to CD33-expressing leukemic cells
- Induces cell death through DNA damage
Contraindications
- Hypersensitivity to gemtuzumab ozogamicin
- Severe hepatic impairment
Adverse Reactions
- Cytopenias (anemia, neutropenia, thrombocytopenia)
- Infusion-related reactions (fever, chills, hypotension)
- Hepatotoxicity (elevated liver enzymes)
- Veno-occlusive disease (VOD)
Drug Interactions
- Hepatotoxic agents (increased risk of liver damage)
- Myelosuppressive agents (increased risk of cytopenias)
