Forms & Strengths
- Gel 1% for topical use
- Capsules 75mg, for oral use
Adult Dosing
- 300mg/m²/day for CTCL
- Initial dose 225-300mg/m²/day, may increase to 400mg/m²/day for refractory or relapsed disease
Pediatric Dosing
- Safety and efficacy not established in patients under 18 years
- Dosing not recommended due to concerns about effects on growth and development
Indications
- Treatment of cutaneous T-cell lymphoma (CTCL)
- Treatment of refractory or relapsed CTCL
Mechanism of Action
- Selective retinoid X receptor (RXR) agonist
- Modulates gene expression, inhibiting cell proliferation and inducing apoptosis
Contraindications
- Hypersensitivity to bexarotene or other retinoids
- Pregnancy, as bexarotene may cause fetal harm
Adverse Reactions
- Headache, asthenia, fatigue
- Hyperlipidemia, hypothyroidism, leukopenia
- Cough, dyspnea, skin reactions
Drug Interactions
- Gemfibrozil: increases bexarotene exposure, may increase risk of adverse reactions
- CYP3A4 inhibitors: may increase bexarotene exposure, monitor for adverse reactions
- Vitamin A supplements: may increase risk of hypervitaminosis A, avoid concurrent use
