Forms & Strengths
- Capsules: 2.3 mg, 3 mg, 4 mg
- Oral administration
Adult Dosing
- Starting dose: 4 mg orally on days 1, 8, and 15 of a 28-day cycle
- Dose adjustment based on toxicity and tolerability
- Concomitant use with lenalidomide and dexamethasone: 4 mg on days 1, 8, and 15
- Concomitant use with rituximab and dexamethasone: 4 mg on days 1, 8, 15, and 22
Pediatric Dosing
- Not established for patients under 18 years of age
Indications
- Multiple myeloma in combination with lenalidomide and dexamethasone for patients who have received at least one prior therapy
- Multiple myeloma in combination with rituximab and dexamethasone for patients who have received at least two prior therapies
Mechanism of Action
- Selective inhibitor of the 20S proteasome
- Induces cell cycle arrest and apoptosis in cancer cells
Contraindications
- Hypersensitivity to ixazomib or any component of the formulation
- Pregnancy and lactation

- Thrombocytopenia
- Anemia
- Neutropenia
- Diarrhea
- Fatigue
- Nausea
- Vomiting
- Peripheral neuropathy
Drug Interactions
- Strong CYP3A inducers: decrease ixazomib exposure
- Strong CYP3A inhibitors: increase ixazomib exposure
- P-glycoprotein substrates: potential for increased toxicity
