- Capsules: 100mg
- Oral formulation
Adult Dosing
- 400mg orally once daily
- Dose reduction to 300mg for intolerable side effects
Pediatric Dosing
- Safety and efficacy not established
- Not recommended for use in pediatric patients
Indications
- Treatment of cutaneous T-cell lymphoma (CTCL)
- Progressive, persistent, or recurrent disease
Mechanism of Action
- Histone deacetylase inhibitor
- Modulates gene expression by increasing acetylation of histones and other proteins
- Induces apoptosis in cancer cells
Contraindications
- Hypersensitivity to vorinostat or its components
- Pregnancy and lactation
Adverse Reactions
- Diarrhea
- Fatigue
- Nausea
- Dysgeusia
- Thrombocytopenia
- Anemia
Drug Interactions
- Coumarin-based anticoagulants: increased INR
- Valproic acid: increased risk of thrombocytopenia and hemorrhage
- Other HDAC inhibitors: increased risk of toxicity
