Forms & Strengths
- Oral tablets: 25 mg
- Oral tablets: 100 mg
Adult Dosing
- 100 mg orally once daily on Days 1-28 in combination with low-dose cytarabine 20 mg subcutaneously twice daily on Days 1-10 of a 28-day cycle
- Continue treatment until disease progression or unacceptable toxicity
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Newly diagnosed acute myeloid leukemia (AML) in patients >=75 years or those with comorbidities precluding intensive induction chemotherapy
Mechanism of Action
- Hedgehog pathway inhibitor that binds to and inhibits Smoothened (SMO), reducing downstream signaling necessary for cancer cell survival and proliferation
Contraindications
- Severe hypersensitivity to glasdegib or any excipients
- Pregnancy and lactation due to severe embryo-fetal toxicity
Adverse Reactions
- Anemia
- Fatigue
- Hemorrhage
- Febrile neutropenia
- Musculoskeletal pain
- Nausea
- Edema
- Thrombocytopenia
- Decreased appetite
- Dysgeusia
- QT prolongation
Drug Interactions
- Strong CYP3A4 inhibitors (may increase glasdegib plasma concentrations)
- Strong CYP3A4 inducers (may decrease glasdegib plasma concentrations)
- QT-prolonging drugs (coadministration increases risk of ventricular arrhythmias)
