talazoparib
L01XK04
Forms & Strengths
- Capsules: 0.25 mg, 0.75 mg, 1 mg
Adult Dosing
- HER2-negative locally advanced or metastatic breast cancer with deleterious or suspected deleterious germline BRCA-mutations: 1 mg orally once daily
- Metastatic castration-resistant prostate cancer (in combination with enzalutamide): 0.5 mg orally once daily
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Mutated germline BRCA-positive, HER2-negative locally advanced or metastatic breast cancer
- Metastatic castration-resistant prostate cancer with homologous recombination repair gene mutations in combination with enzalutamide
Mechanism of Action
- PARP (poly ADP-ribose polymerase) inhibitor (PARP1 and PARP2)
- Traps PARP bound to DNA single-strand breaks, resulting in replication forks stalling, double-strand DNA breaks, and cell death in cells with deficient DNA repair
Contraindications
- Hypersensitivity to talazoparib or any of its excipients
Adverse Reactions
- Fatigue
- Anemia
- Nausea
- Neutropenia
- Thrombocytopenia
- Headache
- Vomiting
- Alopecia
- Decreased appetite
- Dizziness
Drug Interactions
- P-gp inhibitors (e.g., amiodarone, carvedilol, clarithromycin, itraconazole) may increase talazoparib exposure; monitor for toxicity and reduce dose if necessary
- Breast Cancer Resistance Protein (BCRP) inhibitors may increase exposure
