Adult Dosing
- 39 mg orally once daily
- Taken with or without food
- Continue until disease progression or unacceptable toxicity
Pediatric Dosing
- Safety and effectiveness in pediatric patients have not been established
Indications
- Treatment of adult and pediatric patients aged 12 years and older with residual or recurrent grade 2 glioma with a susceptible IDH1 or IDH2 mutation
Mechanism of Action
- Small-molecule dual inhibitor of mutant isocitrate dehydrogenase 1 (IDH1) and IDH2 enzymes
- Reduces levels of the oncometabolite 2-hydroxyglutarate (2-HG)
- Promotes cellular differentiation and reduces proliferation in IDH-mutant glioma cells
Contraindications
- History of severe hypersensitivity to vorasidenib or any of its excipients
Adverse Reactions
- Fatigue
- COVID-19 infection
- Musculoskeletal pain
- Diarrhea
- Nausea
- Headache
- Elevated alanine aminotransferase (ALT)
- Elevated aspartate aminotransferase (AST)
- Seizures
Drug Interactions
- Strong CYP1A2 inhibitors: Avoid coadministration or monitor closely for increased toxicity
- Moderate CYP1A2 inhibitors: Monitor for adverse reactions
- CYP1A2 substrates: Coadministration may alter exposure of sensitive substrates
