Forms & Strengths
- 100mg, 150mg, 200mg, 250mg capsules or tablets
Adult Dosing
- 1000-3000mg daily divided into 3-4 doses for cancer treatment
- 150-450mg daily for infertility in males
Pediatric Dosing
- Not established for patients under 18 years of age
Indications
- Adjunct therapy in the treatment of various solid tumors including renal cell carcinoma and prostatic cancer
- Treatment of male infertility with impaired sperm motility
Mechanism of Action
- Inhibits aerobic glycolysis and decreases mitochondrial respiration, selectively targeting cancer cells
- Antispermatogenic activity by affecting the energy production in sperm cells
Contraindications
- Known hypersensitivity to lonidamine or its excipients
- Severe liver or kidney dysfunction
- Pregnancy and breastfeeding
Adverse Reactions
- Gastrointestinal disturbances including nausea, vomiting, and diarrhea
- Hepatic toxicity with elevated liver enzymes
- Neuromuscular toxicity with myalgia, weakness, and fatigue
- Hormonal changes including decreased testosterone levels
Drug Interactions
- Concomitant use with other chemotherapeutic agents may enhance toxicity
- Cimetidine and other H2-receptor antagonists increase systemic exposure to lonidamine
- Phenobarbital and other enzyme inducers may decrease lonidamine levels
