Forms & Strengths
- injection 1mg/mL
- lyophilized powder for injection 12mg/vial
Adult Dosing
- induction: 0.15mg/kg/day IV until remission or 60 days
- consolidation: 0.15mg/kg/day IV for 25 doses, up to 60 days
Pediatric Dosing
- 0.15mg/kg/day IV until remission, 60 days max
- pediatric dosing is less well established
Indications
- relapsed or refractory acute promyelocytic leukemia (APL)
- newly diagnosed APL in combination with other agents
Mechanism of Action
- promotes apoptosis in cancer cells
- differentially affects cellular pathways in leukemia cells
- induces partial differentiation of APL cells
Contraindications
- pregnancy, especially 1st trimester
- known hypersensitivity to arsenic trioxide
- QT prolongation
Adverse Reactions
- prolonged QT interval
- leukocytosis
- differentiation syndrome
- hyperglycemia
- nausea, vomiting, diarrhea
Drug Interactions
- concomitant use with other QT-prolonging agents
- avoid concomitant use with drugs known to increase arsenic levels
- may decrease levels/effects of: warfarin
