Adult Dosing
- 2.5mg PO qday
- recommended duration of treatment: 5 years
- extended treatment: up to 10 years
Pediatric Dosing
- not approved for pediatric use
- safety and efficacy in pediatric patients not established
Indications
- treatment of hormone receptor-positive breast cancer
- extended adjuvant treatment of early breast cancer
- first-line treatment of advanced breast cancer
- neoadjuvant treatment of breast cancer
Mechanism of Action
- non-steroidal competitive inhibitor of aromatase
- inhibits conversion of androgens to estrogens
- lowers estrogen levels in postmenopausal women
Contraindications
- premenopausal women
- pregnancy
- breastfeeding
- known hypersensitivity to letrozole or any excipients
Adverse Reactions
- hot flashes
- osteoporosis
- nausea and vomiting
- fatigue
- arthralgias
- hair thinning
Drug Interactions
- estrogen-containing products
- tamoxifen
- medications that induce CYP3A4 or CYP2A6
- warfarin
- other aromatase inhibitors
