Adult Dosing
- Advanced breast cancer in postmenopausal women: 2.5 mg orally once daily
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Treatment of advanced breast cancer in postmenopausal women
Mechanism of Action
- Potent, non-steroidal third-generation aromatase inhibitor
- Selectively inhibits the cytochrome P450 enzyme CYP19 (aromatase), preventing conversion of androgens to estrogens
Contraindications
- Known hypersensitivity to vorozole
- Pre-menopausal women
- Pregnancy and lactation
Adverse Reactions
- Hot flashes
- Nausea and vomiting
- Fatigue and asthenia
- Headache
- Arthralgia and myalgia
- Increased sweating
- Weight gain
Drug Interactions
- CYP3A4 inhibitors and inducers may alter vorozole clearance
- Estrogen-containing therapies counteract the pharmacodynamic effects of aromatase inhibition
