Forms & Strengths
- Injectable solution: 50 mg/mL in 1.5 mL ampoules
Adult Dosing
- Refractory acute renal transplant rejection: 0.5 to 1 mg/kg/day administered as an intravenous infusion for 10 to 14 days
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Prevention of acute rejection episodes in renal transplantation
- Treatment of steroid-resistant acute renal transplant rejection
Mechanism of Action
- Suppresses antibody production and T-cell proliferation by inhibiting the differentiation of B-lymphocytes into antibody-producing cells and suppressing cytotoxic T-lymphocyte generation
Contraindications
- Hypersensitivity to gusperimus or its ingredients
- Severe bone marrow suppression
- Active, uncontrolled systemic infections
Adverse Reactions
- Leukopenia
- Thrombocytopenia
- Anemia
- Increased susceptibility to bacterial, viral, and fungal infections
- Nausea and vomiting
- Elevated liver enzymes
Drug Interactions
- Immunosuppressive agents: Increased risk of severe bone marrow suppression and opportunistic infections when combined with other immunosuppressants like azathioprine, cyclosporine, or corticosteroids
- Nephrotoxic drugs: Potential for additive nephrotoxicity when co-administered with agents such as aminoglycosides or amphotericin B
