Forms & Strengths
- Subcutaneous injection: 210 mg/1.5 mL in a single-dose prefilled syringe or autoinjector
Adult Dosing
- Plaque psoriasis: 210 mg subcutaneously at Weeks 0, 1, and 2, then every 2 weeks thereafter
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Treatment of moderate-to-severe plaque psoriasis in adults who are candidates for systemic therapy or phototherapy and have failed or response is inadequate to other systemic therapies
Mechanism of Action
- Human monoclonal IgG2 antibody that binds to the human interleukin-17 receptor A (IL-17RA) subunit, blocking IL-17 cytokine-mediated responses and inhibiting inflammatory cascades
Contraindications
- Crohn's disease
- Severe hypersensitivity to brodalumab or any excipients
- Active, clinically significant infections (e.g., active tuberculosis)
Adverse Reactions
- Suicidal ideation and behavior
- Infections (e.g., oropharyngeal candidiasis, upper respiratory tract infections)
- Arthralgia
- Headache
- Fatigue
- Diarrhea
- Neutropenia
- Injection site reactions
Drug Interactions
- Live vaccines: Avoid concurrent use during brodalumab therapy
- CYP450 substrates: No clinically significant pharmacokinetic interactions expected, but systemic inflammation modulation may alter CYP450 enzyme activity
