Adult Dosing
- Severe alopecia areata: 50 mg orally once daily with or without food
Pediatric Dosing
- Severe alopecia areata (Patients 12 years of age and older): 50 mg orally once daily
Indications
- Treatment of severe alopecia areata in adults and adolescents 12 years of age and older
Mechanism of Action
- Selective inhibitor of Janus kinase 3 (JAK3) and the kinase expressed in hepatocellular carcinoma (TEC) kinase family
- Inhibits signaling of various interleukins and cytokines, reducing the immune-mediated destruction of hair follicles
Contraindications
- Severe active infections, including active tuberculosis
- Known hypersensitivity to ritlecitinib or any of its excipients
- Severe hepatic impairment
Adverse Reactions
- Headache
- Nasopharyngitis
- Upper respiratory tract infection
- Urinary tract infection
- Dizziness
- Rash
- Urticaria
- Folliculitis
- Increased risk of serious infections, malignancy, major adverse cardiac events (MACE), and thrombosis
Drug Interactions
- Strong CYP3A inducers (may decrease ritlecitinib exposure)
- Live vaccines (avoid concomitant use during treatment)
- Strong immunosuppressants (avoid concurrent use due to increased risk of cumulative immunosuppression)
