Forms & Strengths
- Delayed-release oral capsules: 120 mg
- Delayed-release oral capsules: 240 mg
Adult Dosing
- Initial dose: 120 mg orally twice daily for 7 days
- Maintenance dose: 240 mg orally twice daily
- May be taken with or without food; taking with food may reduce flushing
Pediatric Dosing
- Safety and efficacy in pediatric patients (under 18 years) have not been established
Indications
- Treatment of relapsing forms of multiple sclerosis (MS) in adults
- Treatment of moderate-to-severe plaque psoriasis in adults (EU)
Mechanism of Action
- Activates the nuclear factor erythroid 2-related factor 2 (Nrf2) pathway
- Promotes cellular antioxidant response and protects against oxidative stress
- Exerts anti-inflammatory and immunomodulatory effects
Contraindications
- Known hypersensitivity to dimethyl fumarate or excipients
- Concomitant use with other fumaric acid derivative products
Adverse Reactions
- Flushing (warmth, redness, itching, or burning sensation)
- Gastrointestinal events (abdominal pain, diarrhea, nausea, dyspepsia)
- Lymphopenia and decreased white blood cell counts
- Elevated liver transaminases
- Proteinuria
Drug Interactions
- Avoid concurrent use with other systemic immunosuppressive or immunomodulating therapies
- Live vaccines should generally be avoided during treatment
- Nephrotoxic drugs may increase the risk of renal adverse effects
